We've changed the transmission speed of the Profibusnetwork in the hardware configuration of our S7-400 PLC. Do we need to power off the ET200M aswell to reïnitiate.

Yes,

I believe it is the standard procedure for ALL types of Profibus slaves.
Profibus slaves recieves the Profibus parameters from the master upon powerup. One of the parameters is the used baudrate.
