Calibration of pt100

No. O deg C = 100 ohms hence why it is called a PT100 so ambient means nothing

Yes when the equipment is to be calibrated and not the sensor.
No when you want to calibrate a sensor only. (However this is depending on more factors like pressure, medium . Most people use a small heatfurnace with a referencesensor.
I only use ICE water to get them to Zero according to USDA req.
